Recent progresses in fluorescence imaging has enabled development of novel microscopic techniques such as superresolution imaging. These techniques demand methods for biomoleuclar labeling using fluorescent probes whose fluorescence is regulated at will for application to the microscopic techniques. In this study, we develop and improve a protein labeling technique based on PYP-tag and its synthetic fluorescent probes. Furthermore, we also design and develop imaging probes with fluorescence switches. Using these chemical tools, we aim at imaging localization and translocation of proteins with high spatiotemporal resolution.
