Development and application of the multimodal two-photon microscopy using the SHG-specific dye

To simultaneously visualize and analyze multiple biological phenomena, we are going to develop the true multimodal two-photon imaging technique that capitalize distinct optical phenomena and apply it to biology. By using the newly developed SHG (second harmonic generation)-specific dye, SHG imaging of the membrane structure and physiology will be established. This will be followed by its simultaneous imaging of SHG with various fluorescent reporters of chemical changes near the membrane. After establishing the widely applicable imaging system that allows investigations of juxtamembrane phenomena from multiple aspects, it will be applied to brain samples to reveal information processing of neurons and glial cells.
